Abstract:

Background: In this study we evaluate the effects of different doses of GDF9 with fixed dose of BMP15 on quality of in vitro maturated sheep oocytes. Materials and Methods: Good ovine Cumulus-oocyte complexes (COCs) were first transferred to Pre-IVM medium, and then to three IVM mediums supplemented with 10%v/v BMP15 and: 1) 175 ng/ml 2) 131 ng/ml or 3) 219 ng/ml GDF9. After maturation period, MII oocytes with normal first polar body were counted. Oocyte viability was determined by means of the TUNEL technique. Immunofluorescence microscopy staining were used to evaluate cell meiotic spindle and redistribution of cortical granules (CGs). In addition, fertility rate in group 1 and 2 was determined by means of IVF. Statistical analyses were carried out by ANOVA and significant differences between means were determined using Tukey HSD test for comparison of multiple means. Results: Maturation rate of group 3 was significantly lower than group 1 and 2 (75.57% ± 2.74 vs. 93/56% ± 0.56 and 90.67% ± 4.09, respectively, p<0.05). Spindle,CGs, and TUNEL staining results indicated no significant difference between groups. MII oocytes with normal polar body in group 1 was significantly higher than group 2 and 3 (71.39% ± 0.75 vs. 25.80% ± 1.09 and 31.68% ± 2.36, respectively, p<0.001). Finally, the blastocyst formation in group 1 was significantly higher than group 2 (39.86% ± 0.38 vs. 28.73% ± 1.77, p<0.01). Conclusion: These results demonstrate that SPOM medium supplemented with 175 ng/ml GDF9 and 10% BMP15 enhance oocyte developmental competence.

P-136: In Vitro Development of Ovine Oocytes Matured in A Chemically Defined Medium

Background: Despite great advances achieved in the field of in vitro oocyte maturation (IVM), almost all IVM media are supplemented with undefined component which not only pose the risk of pathogen transmission but also hinders our knowledge of molecular mechanisms of maturation. One of these components is serum that containing various concentration of unknown molecules affecting cellular proce...

P-118: Effect of Vitrification on Developmental Competence of Parthenogenetic Activation in In Vitro Matured Ovine Oocytes

Background: Cryopreservation of in vitro matured oocytes is a useful technique because the oocytes can be used for some assisted reproductive technologies. On the other hand, the cryopreservation of oocytes is an open problem as a result of their structural sensitivity to the freezing process. The purpose of this study was to evaluate the effect of vitrification on in vitro development of vitri...

P-110: Effect of Increasing Amount of Oocyte Secreted Factors on Cumulus Expansion of Bovine Cumulus-Oocyte Complexes

Background: In vitro maturation is a good method to decrease cancer risk of superovulation by gonadotropin hormones. A paracrine effect of oocyte secretions on oocyte developmental competence is under investigation. Apart from oocyte maturation, ovulation in vivo requires a precise control of extracellular matrix modification. Cumulus cells secrete hyaluronan to form a muco-elastic extracellula...

P-24: The Effect of Oocyte Activation with DTT on Development of Ovine Embryos in ICSI

Background: The objective of the current study was to determine the effect of activation of injected oocytes with dithiothreitol on development of ovine embryos derived from in vitro-matured oocytes and intracytoplasmic sperm. The injected oocytes with non-treated sperm were activated with 1) ionomycin (Io) + 6-dimethylaminopurine (6-DMAP) 2) DTT, 3) DTT+ ionomycin (I)+6-DMAP. P-64: Anthocyanin Effects on Sheep Oocytes In Vitro Maturation in The Presence of cAMP Modulators.

Background We have previously reported the effect of Anthocyanin and cAMP modulators on maturation and apoptosis of sheep oocytes. In this study, we investigated the reduction capacity and apoptotic gene expression results at MII sheep oocytes. MaterialsAndMethods Good ovine Cumulus-oocyte complexes (COCs) were transferred to Pre-IVM medium, then to two type IVM mediums: 1) 0.1μg/ml Delphinidin...

P-119: Comparison of Weymouth and TCM 199 for Ovine Oocyte Maturation

Background: Different culture media are used for in vitro maturation of ovine oocyte. The Weymouth medium are used for in vitro maturation of rat and mice oocytes with very low levels of supplementation (Just FBS, while to acquire the good results in TCM hormone, and serum supplementation and in some cases growth factor are necessary to gain the optimized results. The aim of the present study w...
